產(chǎn)品簡(jiǎn)介:

gus(β-glucuronidase,β-D- 葡萄糖苷酸酶)基因是目前常用的一種報告基因,其表達產(chǎn)物β-葡萄糖苷酸酶 (GUS)是一種水解酶,能催化許多β-葡萄糖苷酯類(lèi)物質(zhì)的水解,它可以將5-溴-4-氯-3-吲哚-β-葡萄糖苷酸酯(x-gluc)分解為藍色的物質(zhì),其檢測方法簡(jiǎn)單、快速、靈敏、穩定,且背景活性低。因為絕大多數植物細胞內不存在內源的GUS活性,因此gus基因廣泛用作轉基因植物的報告基因,尤其是在研究外源基因瞬時(shí)表達的轉化實(shí)驗中廣泛應用。

該試劑盒包含GUS染色的全部試劑,使用方便,只需將配制好的X-gluc溶液和緩沖液按照比例混合即配成GUS染色液。該試劑盒可以配制50ml GUS染色液。

貯存和效期:

未配制的試劑盒按照溫度貯存,有效期2年。

配制好的X-gluc溶液-20℃避光保存30天,長(cháng)期-80℃保存;GUS染色緩沖液 4貯存;GUS染色工作液最好現用現配,短期貯存可以-20℃保存2-3

使用說(shuō)明:

一、X-gluc溶液(50×)配制:

吸取1ml X-gluc溶解液加入到X-gluc管中,徹底混勻,至粉末完全溶解,即配成X-gluc溶液(50×),該溶液-20℃避光保存。

注:X-gluc溶液穩定性較差,-20℃避光保存30天左右溶液保持無(wú)色,長(cháng)期保存建議按照需要分裝到棕色管里,-80℃避光保存。正常的X-gluc溶液顏色為無(wú)色,如果溶液變?yōu)榧t色或棕色,表明溶液失效。

注:GUS染色工作液最好現用現配,短期貯存可以-20℃保存2-3天。

. GUS染色步驟:

1.取材:將葉片、花瓣、根莖等組織剪成小片,放于1.5ml離心管中。

注:用于染色的植物材料的制備方法要因涉及的特定組織和器官的不同而異。例如,擬南芥的根、花和葉片以及煙草幼苗的根就可以不作任何預處理而直接染色。但是像煙草和馬鈴薯這些植物的莖和葉就必須在染色前切成薄片(1-3mm)。當操作大的組織和樣品時(shí),可以選用真空滲入法來(lái)幫助底物滲入細胞。

2.染色:加入適量配制好的GUS染色工作液至完全覆蓋材料;錫箔紙包好后搖床搖動(dòng)10分鐘或更長(cháng)時(shí)間至材料出現藍色。注意要做陰性和陽(yáng)性對照實(shí)驗。 

3.洗脫:將材料轉入無(wú)水乙醇或80%丙酮中脫色2-3次,至陰性對照材料為白色。GUS染色陽(yáng)性的藍色斑點(diǎn)很穩定,在酒精中不褪色。

4.觀(guān)察:一般染色后,GUS染色陽(yáng)性的藍色斑點(diǎn)肉眼就能看到。但是有些材料可能藍色斑點(diǎn)很細微,要在顯微鏡下才能看到。
